Resumo: As respostas de células eucarióticas frente a agentes endógenos e exógenos que causam danos no DNA (genotóxicos) dependem do acionamento rápido e eficiente de vias de sinalização e/ou redes integradas de sinalização que levam ao reparo do material genético lesado e a manutenção da estabilidade/integridade do mesmo possibilitando a sobrevivência e proliferação celular. Estas respostas são mediadas por mecanismos de Transdução de Sinais, muitos dos quais envolvem as famílias de enzimas Rho GTPases e Proteínas Tirosina Fosfatases e Quinases.

Assim sendo, a Regulação do Ciclo Celular e de Mecanismos Moleculares de Manutenção da Estabilidade Genômica de células eucariótics são os principais interesses de nosso laboratório em diferentes contextos fisiopatológicos. Para isso, uma ampla gama de abordagens Bioquímicas, de Proteômica, de Biologia Celular e Molecular, de Biologia de Sistemas e Bioinformática (Interação Proteína-Proteína ou Interactomas) são usadas para se investigar as funções moleculares e celulares destas enzimas.  Os modelos biológicos que usamos em nossos estudos compreendem linhagens celulares normais e tumorais humanas (melanoma, glioma, câncer de cervix, mama, pulmão e próstata, leucemias) a agentes externos (físicos e químicos) causadores de danos no DNA genômico que mimetizam terapias e tratamentos anticâncer. 

Um maior entendimento e conhecimento de redes de sinalização intracelular controladas por estas enzimas nos levarão ao melhor planejamento e intervenção molecular por pequensa moléculas (drogas) que possam viabilizar tratamentos terapêuticos, usando pré-sensibilizações combinadas de quimioterápicos associadas ou não com radioterapia, contra os diferentes tipos de câncer humanos de maior incidência na população.

Introduction
ABSTRACT: The responses of eukaryotic cells to endogenous and exogenous agents that cause DNA damage (genotoxic) depend on the fast and efficient activation of signaling pathways and/or integrated signaling networks that lead to the repair of lesioned genetic material and maintenance of genetic stability/integrity enabling cell survival and proliferation. These responses are mediated by Signal Transduction mechanisms, many of which involve the Rho GTPase, Protein Tyrosine Phosphatases and Kinase families of enzymes.
 
Therefore, Cell Cycle Regulation and Molecular Mechanisms for the Maintenance of Genomic Stability of eukaryotic cells are the main interests of our laboratory in different pathophysiological contexts. For these purposes, a wide range of Biochemical, Proteomics, Cellular and Molecular Biology, Systems Biology and Bioinformatics (Protein-Protein Interaction or Interactomes) approaches are used to investigate the molecular and cellular functions of these enzymes. The biological models we use in our studies range from human normal and tumor cell lines (melanoma, glioma, cancer of the cervix, breast, lung and prostate, leukemia) to external agents (physical and chemical) causing damage to genomic DNA that mimic therapies and anti-cancer treatments.
 
A greater understanding and knowledge of intracellular signaling networks controlled by these enzymes will lead us to better planning molecular interventions by small molecules (drugs) that can enable therapeutic treatments, using combined pre-sensitization of chemotherapy agents associated or not with radiotherapy, against the different types of human cancers with the highest incidence in the population.
